The Uses of FACTOR XA

Fusion proteins are commonly expressed with a factor Xa cleavable Ile-Glu (or Asp)-Gly-Arg-↓-X sequence. Typically 1 mg of fusion protein can be incubated with 10 μg of factor Xa for 2.5 hours at 37 °C.

General Description

Native coagulation factor Xa from human plasma prepared from homogeneous Factor X by activation with Russel’s viper venom. Hydrolyzes peptide and ester bonds at the carboxyl group of arginine. Specifically catalyzes the activation of prothrombin to thrombin. Prothrombin activation is accelerated by Factor Va, phospholipids, and Ca²⁺. Factor Xa was prepared from homogeneous Factor X, using Russels′ viper venom (RVV), which was removed after activation. Inhibited by AEBSF (Cat. No. 101500), DFP (Cat. No. 30967), PMSF (Cat. No. 52332), and soybean trypsin inhibitor (Cat. No. 650357). Sold on the basis of plasma equivalent units of Factor X. 100 plasma equivalent units are equal to 800 μg of Factor X.

Biochem/physiol Actions

Factor Xa catalyzes the hydrolysis of the Arg-Thr and then Arg-Ile bonds in prothrombin to yield active thrombin.The fairly strict recognition sequence is Ile-Glu (or Asp)-Gly-Arg-↓-X.It may sometimes cleave at other basic residues, depending on the conformation of the target protein. Factor Xa will not cleave if a proline residue follows the arginine of the recognition sequence.pH Optimum: 7.6-8.0Temperature Optimum: 37 °C
