General Description

CoREST (also referred to as RCOR1 (REST corepressor 1)) mRNA is ubiquitously expressed in all tissues. It is a corepressor of the RE1-silencing transcription factor REST. In nonneuronal cells, the transcription factor binds to RE1 neuron-restrictive silencer elements and suppresses gene expression. CoREST is a component of the BRAF35 (BRCA2-associated factor 35)–histone deacetylase complex. The RCOR1 gene is mapped to human chromosome 14q32.
Human recombinant CoREST, GenBank Accession No. NM_015156), amino acids 305-end, with N-terminal His tag, MW = 20kDa, expressed in Escherichia coli expression system.

Biochem/physiol Actions

CoREST associates with REST (RE1-silencing transcription factor), the histone deacetylases 1 and 2 (HDAC1/2), and LSD1 (lysine-specific histone demethylase 1A). This complex is involved in chromatin remodeling for changing gene expression. The LSD1/CoREST/REST compressor complex is also required for the expression of Herpes Simplex virus genes. LSD1 along with CoREST is responsible for the oxidative demethylation of the H3 histone (mono- and di-methylated Lysine4 residue). Deletions in the CoREST gene results in unfavorable clinical outcomes in DLBCL (diffuse large B-cell lymphoma) patients. It might also participate in the epigenetic control of blood cell development.
